Senescence causes age-related diseases and stress-related injury. Paradoxically, it is also essential for organismal development. Whether senescence contributes to lung development or injury in early life remains unclear. This talk will discuss the emerging role of senescence in modulating normal lung development and injury. Our work demonstrates that 1) lung senescence occurs at birth and decreases throughout the saccular stage in mice. This contributes to normal lung development; 2) neonatal hyperoxia interrupts the normal reduction in developmental senescence by causing a transient increase during the alveolar stage in mice; 3) single-cell RNA sequencing data show that this senescence occurs in macrophages, type II cells and secondary crest myofibroblasts; 4) removing senescent cells by senolytics during the alveolar stage attenuates hyperoxia-induced alveolar and vascular simplification. Conclusively, early programmed senescence orchestrates normal lung development whereas later hyperoxia-induced senescence causes lung injury. This defines the ontogeny of lung senescence and provides an optimal therapeutic window for mitigating neonatal hyperoxic lung injury by inhibiting senescence.

姚宏伟，教授，一直从事慢性肺疾病的分子病理机制和治疗靶点的研究。共发表文章150余篇，其中SCI论文100余篇，包括以第一或通讯作者身份发表在Nature Communications, Journal of Clinical Investigation和PNAS杂志上。总引用次数达6000以上，H指数为45。主持美国NIH/R01基金，美国杰出生物医学研究中心 (COBRE) 基金子项目，国家自然科学基金以及Ralph博士和Marian Falk医学研究信托基金等多项基金。

2004年获得中国药理学会-法国施维雅青年药理学工作者奖（每年全国只有8名）。2008年获得安徽省科学技术奖三等奖。2011年获得罗切斯特大学最佳论文发表奖。2022年获得美国胸科学会Whitsett奖。2022年主持NIH workshop：“线粒体与肺疾病和睡眠失调病理机制”。特约为包括美国NIH基金，英国MRC基金和国家自然科学基金以及七十几种期刊杂志评审近千次。2022年被遴选为中国医学科学院学部委员选聘工作同行评议专家。担任Frontiers in Pharmacology杂志副主编，American Journal of Physiology - Lung Cellular and Molecular Physiology，Frontiers in Bioscience和Physiological Reports 等多种国际杂志编委。
